Purification of Alkaline Phosphatase

چکیده
8. An increase in the rate of progression of the bands down the column decreases the sharpness of the bands. On 'Zeo-Karb 215' (40-60 mesh/in.) a rate ofprogression of 10-15 cm./hr. gave satisfactory results. 9. Equations have been derived permitting the calculation ofthe proportion ofthe column occupied by a component, the width ofthe boundaries and the expected yield of pure components in separation experiments. 10. Details of a specimen separation are given, and the yields obtained compared with estimates derived from theory.

P-178: Separation and Identification of Alkaline Phosphatase Isozymes during Pregnancy
Background: Alkaline phosphatase (ALP), (EC 3.1.3.1) is a hydrolase enzyme responsible for removing phosphate groups from various molecules in the body. In humans, ALPis present in all tissues such as liver, bile duct, kidney, bone, and the placenta which detection of its activity is so useful in molecular biology. Pregnancy is associated with normal physiological changes that assist fetal surv...
متن کاملIsolation and purification of placental type alkaline phosphatase from a seminoma.
An alkaline phosphatase (EC 3.1.3.1) of the placental type was isolated from a seminoma type of human testicular cancer tissue and was purified to homogeneity by sulfate-mediated chromatography on a column of Cibacron Blue Sepharose 4B. The purified enzyme had a specific activity of 40.6 kU per gram of protein and was obtained in a yield of 37%. The purification procedure used was simple and ec...
متن کاملRunning title: Alkaline phosphatase inhibition and Francisella survival 1 Francisella DnaK Inhibits Tissue Nonspecific Alkaline Phosphatase
Following pulmonary infection with Francisella tularensis, we observed an unexpected but significant reduction of alkaline phosphatase, an enzyme normally up-regulated following inflammation. However, no reduction was observed in mice infected with a closely related Gram negative pneumonic organism (Klebsiella pneumonia) suggesting the inhibition may be Francisella specific. In similar fashion ...
متن کاملSeparation and Characterization of Alkaline Phosphatase Isoenzymes from Blood and Other Tissues
Alkaline phosphatases may be contributed to the blood by a number of organs; thus the serum alkaline phosphatase may consist of a mixture of phosphatase isoenzymes. Whether or not these isoenzymes differ other than in their tissues of origin has long been debated, some evidence indicating that they are identical, some that they are different. Attempts to characterize alkaline phosphatase by a s...
متن کاملAlkaline inorganic pyrophosphatase activity of mammalian-cell alkaline phosphatase.
Alkaline phosphatase prepared from mammalian cell cultures was found to have alkaline inorganic pyrophosphatase activity. Both of these activities appear to be associated with a single protein, as demonstrated by: (1) concomitant purification of alkaline phosphatase and alkaline inorganic pyrophosphatase; (2) proportional precipitation of alkaline phosphatase and inorganic pyrophosphatase activ...
